That's not possible. If a gfx card which doesn't support 32 bit mouse pointers (all but radeon gfx cards) is used and the SOFTSPRITE tooltype isn't set either intuition ignores the 32 bit mouse pointers and uses 2 (on very few gfx cards 3) colour pointers instead, but there is no way to check if 32 bit pointers are supported.
